Mendes är av kubansk börd. Sedan 2011 har hon en relation med skådespelaren Ryan Gosling som hon lärde känna när de spelade mot varandra i The Place Beyond the Pines. Tillsammans har de två döttrar, födda 2014 och 2016.

Gosling is a supporter of People for the Ethical Treatment of Animals (PETA), Invisible Children, and the Enough Project and has traveled to Chad, Uganda and eastern Congo to raise awareness about conflicts in the regions. He has been involved in peace promotion efforts in Africa for over a decade.
